A good way to begin the analysis is to use a good example of real mtDNA. Not long ago I obtained a complete genome sequencing (WGS) from Dante Labs with a particular obtain mtDNA sequencing that cost only a few hundred dollars. Going forward, it’ll be important to analyze mtDNA from compartments other than saliva to get a much better handle on real heteroplasmy within different mitochondria. For instance, cardiac muscle cells or fibroblasts can collect mutated and erased mtDNA preferentially. In other cases, different tissues deliberately maintain separate populations of heteroplasmic mitochondria.

The good news, here, is that without everyone is successful in the potluck of hereditary recombination, everyone with mitochondria can be assigned to a particular haplotype.
